The following are recipes and MSDS sheets for components of the reagents used in the WSSP research project.

Recipes for WSSP reagents:

LB  Add the following to 800ml H2O, 10g Bacto-tryptone.  (1% final); 5g yeast extract. (0.5% final); 10g NaCl. (1% final)
            Adjust pH to 7.5 with NaOH.   Adjust volume to 1L with dH2O.  Sterilize by autoclaving

Miniprep Soln I: 100 mM Tris-HCl (pH 7.5); 10 mM EDTA

Miniprep Soln II: 0.2 N NaOH, 1% SDS

Miniprep Soln III: 0.3 M Na acetate

Miniprep Wash Buffer: 80% Ethanol

MiniprepElution Buffer: 10 mM Tris-HCl (ph 7.5),  1 mM EDTA

TAE Gel Buffer (http://biotech.about.com/od/buffersandmedia/ht/TAE.htm)

Make a concentrated (50x) stock solution of TAE by weighing out 242 g Tris base (FW = 121.14) and dissolving in approximately 750 mL deionized water. Carefully add 57.1 ml glacial acid acid and 100 mL of 0.5 M EDTA (pH 8.0) and adjust the solution to a final volume of 1 L. This stock solution can be stored at room temperature. The pH of this buffer is not adjusted and should be about 8.5.
